Glycolysis by Sarcoma 180 of mice treated with 6-mercaptopurine. I. The role of various enzymes in lactate production.

چکیده
Lactate formation from fructose-1,6-diphosphate in homogenates of Sarcoma 180 from mice, given injections intraperitoneally of 6-mercaptopurine, was sharply de creased as compared with that in tumor homogenates of nontreated animals. The activities of aldolase and triosephosphate isomerase, but not of phosphoglyceraldehyde dehydrogenase and phosphoglyceric acid kinase, were significantly lower, and the ATPase activity was markedly higher in the tumor homogenates of the 6-mercaptopurine-treated animals. The phosphate balance in the glycolysates of this group of homogenates was always negative and was not changed by the addition of glucose to the medium. Addition of crystalline aldolase increased the rate of lactate output to the level of that of homogenates of the nontreated group and shifted the phosphate balance to positive values. Addition of crystalline lactic dehydrogenase or of glucose to either group of homogenates did not significantly change their lactate output.

Enzyme, nitrogen, and DNA concentrations in sarcoma 180 in mice treated with 6-mercaptopurine.
The concentrations of homogenate-nitrogen, of DNA, and the activities of various nucleotidases, adenosine deaminase, cathepsin and lactic dehydrogenase have been studied in homogenates of Sarcoma 180 from non treated mice, from mice given injec tions of 6-carboxymethylcellulose in saline, and from mice treated with 6-mercaptopurine suspended in saline containing 6-carboxymethylcellulose. Statis...
متن کاملEffect of selected tumor-inhibitory agents on the transplantability of sarcoma 180 ascites.
Several compounds known to inhibit the growth of Sarcoma 180 were compared for their cytocidal activity in vivo. Standard inocula from donor mice which were treated with massive doses of the drugs were transplanted at 4 and 24 hours after treatment into untreated recipient animals. Viability was evaluated by cell counts and survival time. The effectiveness of triethylenethiophosphoramide and ac...

متن کاملAerobic Glycolysis in the Frontal Cortex Correlates with Memory Performance in Wild-Type Mice But Not the APP/PS1 Mouse Model of Cerebral Amyloidosis.
UNLABELLED Aerobic glycolysis and lactate production in the brain plays a key role in memory, yet the role of this metabolism in the cognitive decline associated with Alzheimer's disease (AD) remains poorly understood. Here we examined the relationship between cerebral lactate levels and memory performance in an APP/PS1 mouse model of AD, which progressively accumulates amyloid-β. In vivo (1)H-...
متن کاملAlkaline phosphatase activities of 6-thiopurine-sensitive and -resistant sublines of Sarcoma 180.
The alkaline phosphatase activities of 6-thiopurine-sensitive and -resistant sublines of Sarcoma 180 were exam ined in a further study of these enzymes, which have a role in the resistance of Sarcoma 180/TG to 6-thioguanine and 6-mercaptopurine. Both neoplasms possessed particulate-bound alkaline phosphatase enzymes labeled A and B on the basis of their ease of solubilization from the particula...
